Fosaprepitant dimeglumine has four functional groups which have pKa values of 3.05 ± 0.03, 4.92 ± 0.02, 9.67 ± 0.01 and 10.59 ± 0.03. The pka value of 3.05 corresponds to the morpholinium group, the pka of 4.92 corresponds to the monophosphonate group, the pka of 9.67 corresponds to the meglumine counter ion, and the pka of 10.59 corresponds to the triazolinone NH group.

福沙吡坦二甲葡胺性质、用途与生产工艺
神经激肽受体有NK1、NK2和NK3三种亚型。其中NK1受体与P物质的结合能力最强，在大脑呕吐中枢中含量最高。NK1受体与中枢P物质结合后激发呕吐、焦虑、偏头痛等一系列病理生理过程。阿瑞匹坦、福沙匹坦等NK1ＲAs通过选择性与NK1受体结合，有效防治CINV。通常急性期CINV与外周5-HT3能通路活性有关，而迟发性CINV而与中枢P物质有关，因此，与5-HT3ＲAs相比，阿瑞匹坦和福沙匹坦对于迟发性呕吐显示了明显的优势。
福沙吡坦二甲葡胺是Merck公司研发的神经激肽-1受体拮抗药(neurokinin-1receptorantagonist，NK-1RA)，2008年1月首次在美国上市。福沙匹坦二甲葡胺是第一个NK-1RA药物阿瑞匹坦(aprepitant)的前体药物，进入体内后可迅速转化为阿瑞匹坦，临床用于防治中、高度致吐性化疗(包括高剂量的顺铂)初始和反复用药引起的急性或延迟性恶心与呕吐。

Fosaprepitant (MK-0517, L-758,298)是一种aprepitant的磷酰基前药。Aprepitant是一种选择性P物质(NK-1 受体)拮抗剂，被批准用作与皮质类固醇和 5-HT 3受体拮抗剂的联合疗法，以预防急性和延迟化疗诱导的恶心和呕吐。
Fosaprepitant静脉内给药30分钟内通过常见磷酸酶的作用转化为aprepitant。Fosaprepitant在高达150 mg (1 mg/ml)剂量下耐受性良好，Fosaprepitant 115 mg在其AUC中生物等效于aprepitant 125 mg。Fosaprepitant 115 mg已经被提交到FDA，作为aprepitant口服给药3天疗程里第1天代替给药，aprepitant在第2和第3天口服给药。
