辛伐他汀内酯二醇性质、用途与生产工艺
莫那可林J是合成心血管药物辛伐他汀的主要前体。其具有内脂形式(辛伐他汀内酯二醇)和三羟基酸两种形式,合成辛伐他汀的路线主要有两种:一是通过化学法直接将洛伐他汀甲基化得到辛伐他汀;二是是通过化学或生物方法将2,2‑二甲基丁酰基结合到莫那可林JC8位的羟基得到辛伐他汀。美国专利报道的方法:通过LovD表达的酰基转移酶可以高效、专一地将2,2‑二甲基丁酰基结合到莫那可林JC8位的羟基得到辛伐他汀,该工艺简单,反应条件温和,有工业化生产的前景。莫那可林J作为合成心血管药物辛伐他汀的主要前体,其制备工艺也决定着辛伐他汀的生产成本。目前,就莫那可林J合成辛伐他汀的技术路线中,都是以洛伐他汀纯品为原料进行碱水解得到莫那可林J。
Monacolin J 是胆固醇合成抑制剂,能够抑制 HMG-CoA reductase 的活性。
Monacolin J is an inhibitor of cholesterol biosynthesis, and inhibits the activity of HMG-CoA reductase. Na salt of Monacolin J causes 50% inhibition on reductase at 1.2 μg/mL.
