Step 1:
A mixture of 1H-indazole-5-carbaldehyde 15a (5.0 g, 34.25 mmol), sodium hydroxide (4.1 g, 102.7 mmol), iodine (17.4 g, 68.5 mmol) and 10 mL DMF was stirred overnight at room temperature.
The reaction solution was concentrated under reduced pressure, and the residue was purified by silica gel column chromatography to obtain compound 3-iodo-1H-indazole-5-carbaldehyde 15b (9.1 g, yellow solid) with a yield of 97%. MS m/z (ESI): 273.1 [M+1].
