4.2 General procedure for the synthesis of 2-(3-pyridyl)/2-amino thiazole derivatives (3/5)
General procedure: A mixture of phenacyl bromide (1a) (10.0mmol), pyridine-3-carbothioamide/thiourea (2/4) (10.0mmol) and 5.0wt% of silica-supported HClO4 was taken in a mortar and ground together with a pestle for 5-10min at RT. At this stage, the progress of the reaction was monitored by TLC. After completion of the reaction, the crude mass was washed with water (20mL) twice and extracted with ethyl acetate (10mL) twice. Then the catalyst was separated by filtration under vacuum. The organic layer was washed with water, dried with anhydrous Na2SO4 and the solvent was evaporated under reduced pressure. The obtained product (3a/5a) was purified by recrystallization using ethanol. The same procedure was applied for the preparation of all other compounds (3b-k & 5b-k). All the compounds gave satisfactory physical and spectroscopic data following their proposed procedure. Compounds 5a-d and 5f-5k were synthesized and reported in the literature [82,83].
