Reaction Conditions:

with phenoxyamine hydrochloride in methanol;aq. phosphate buffer;water-d2; pH=6.5 at 60; for 8 h;

Steps:

1.1.17 General procedure for the formation of nitriles
General procedure: To a 7mL vial, aldehyde (1 equiv) and O-phenylhydroxylamine hydrochloride (H2NOPh) (1.2 equiv) were dissolved in 4:1 of methanol:0.5M sodium phosphate pH 6.5 to the final concentration of 0.1M. The reaction was stirred at 60°C until the changes of starting materials could not be observed (monitored by TLC). Then, the reaction mixture was cooled to room temperature and extracted with 3mL CH2Cl2 a total of 5 times. The combined organic layer was dried over Na2SO4 and concentrated by rotary evaporator. The crude product was purified by silica column chromatography.
