Example B-8: Preparation of (2S)-1-(4-(1-isopropyl-3-(2-methylimidazo[1,2-a]pyrimidin-6-yl)-1H-pyrazol-4-yl)pyrimidin-2-ylamino)propan-2-ol (B-8). Preparation of 6-bromo-2-methylimidazo[1,2-a]pyrimidine (B-8-2). To a mixed solution of 5-bromopyrimidin-2-amine (170.0 g, 0.98 mol) in isopropanol (2 L) and N,N-dimethylformamide (500 mL) was added 1-chloro-2-propanone (606 g, 6.53 mol). The reaction mixture was stirred at 100 °C for 16 hours. Completion of the reaction was confirmed by monitoring the progress of the reaction by thin layer chromatography (TLC) (unfolding agent ratio: petroleum ether/ethyl acetate = 1:2). After completion of the reaction, the reaction was quenched with sodium carbonate (Na2CO3) solution and filtered. The filtrate was concentrated under reduced pressure and the residue was purified by column chromatography (eluent ratio: petroleum ether/ethyl acetate=5:1) to afford the target compound B-8-2 (10.0 g, 4.8% yield) as a yellow solid. | [References]
