Methylacetophos was produced commercially through a streamlined organophosphate route in which manufacturers first generated the O-methyl phosphoramidate portion and then coupled it to an acetophenone derived oxime. Industrial processes typically began with dimethyl phosphoramidic dichloride, which was reacted with methanol and an amine to form the appropriate phosphoramidate intermediate. In parallel, p-nitroacetophenone was converted to its oxime, then reduced and activated so it could undergo O-phosphorylation with the phosphoramidate reagent. The final step produced the characteristic phosphoramidothioate structure of methylacetophos, followed by purification and formulation. | [Mechanism of action]
