General procedure for the synthesis of methyl 2-(2-amino-5-methyl-4-thiazolyl)acetate from the compound (CAS: 17356-08-0) and methyl 4-bromo-3-oxopentanoate: To a solution of bromide (18 g, 86 mmol) in toluene (100 mL) prepared in step 1 was added thiourea (10.5 g, 138 mmol). The reaction mixture was heated to 100 °C and held for 1 h. Subsequently, it was cooled to room temperature and the solvent was removed under reduced pressure. The residue was dissolved in dichloromethane (100 mL), saturated sodium bicarbonate solution (75 mL) was added and the mixture was stirred vigorously for 10 minutes. The organic layer was separated, dried over anhydrous sodium sulfate, filtered, and concentrated under reduced pressure. Subsequently, the residue was recrystallized by dichloromethane/hexane solvent mixture to give the target product (10 g, 63% yield) as a white solid. The product (C7H10N2O2S) was characterized by the following data: LC-MS, retention time 0.76 min, (M+H)+ m/z 187.0; 1H NMR (CDCl3): δ 2.23 (s, 3H), 3.70 (s, 2H), 3.75 (s, 3H), 4.83-4.95 (wide s, 2H). | [References]
