Step 2: 2-(Biphenyl-4-yl)-N-methoxy-N-methylacetamide (500 mg, 1.96 mmol) was dissolved in anhydrous tetrahydrofuran under nitrogen atmosphere and cooled to -78 °C. Subsequently, phenylmagnesium bromide solution (5 mL, 5 mmol) was slowly added and the reaction was stirred for 3 hours keeping the temperature at -78 °C. The progress of the reaction was monitored by thin layer chromatography (TLC) and after confirming that the starting material was completely consumed, the reaction mixture was diluted with ethyl acetate and washed with water and saturated saline in turn. The organic phase was dried over anhydrous sodium sulfate and concentrated under reduced pressure, and the resulting crude product was purified by column chromatography (petroleum ether: ethyl acetate = 20:1) to afford Intermediate 5 (430 mg, 80.7% yield).
