2-Amino-5-bromopyrazine (1,170 mg, 6.72 mmol) was dissolved in tetrahydrofuran (THF, 100 mL) and iodomethane (CH3I, 1.2 mL, 19.5 mmol) was added. The reaction mixture was cooled to 0 °C, 60% sodium hydride (NaH, 1.5 g, 37.5 mmol) was added slowly and stirred at this temperature for 5 hours. Upon completion of the reaction, a small amount of methanol was added to quench the excess sodium hydride, followed by the addition of water (10 mL). The reaction mixture was extracted with ethyl acetate (3 x 200 mL), the organic layers were combined and dried over anhydrous sodium sulfate. The organic layer was concentrated under reduced pressure and the resulting crude product was purified by column chromatography [silica gel 48.5 g; eluent: hexane-ethyl acetate (1:1)] to afford 5-bromo-2-(dimethylamino)pyrazine (1,096 mg, 5.4 mmol, 80% yield) as a light yellow powdery solid. | [References]
