3-(Trifluoromethyl)-[1,2,4]triazolo[4,3-a]pyrazine can be used as reactant/reagent in synthetic preparation of 3-amino-4-phenylbutanoic acid derivatives as dipeptidyl peptidase inhibitors for treatment or prevention of diabetes. | [Synthesis]
General method: 10 mmol of compound 3 (CAS:1447800-65-8) was dissolved in 50 mL of toluene and 20 mmol of triethylamine was slowly added to the reaction system at room temperature with continuous stirring for 1 hour. The reaction process was monitored by thin layer chromatography (TLC). After the reaction was complete, the reaction mixture was slowly poured into 25 g of ice-water mixture, stirred thoroughly and left to stratify. The organic phase was separated and washed with 2 x 10 mL of cold water, followed by drying the organic phase with anhydrous sodium sulfate. Finally, the organic phase was concentrated under reduced pressure to give compound 4 in good yield. | [References]
