(1)色谱条件 色谱柱:载体,Chromsorb W-HP (80~100目);固定液,3%FFAP(聚乙二醇2mol/L和2-硝基苯甲酸的反应物);(2m×2mm)不锈钢柱;柱温:158℃;进样器温度;200℃;检测器:FID;载气:N2流速30ml/min;H2流速0.6ml/min;空气压力:49.03kPa。
(2)对照品及内标溶液的配制 精密称取对照品11.11mg,置于1ml量瓶中,加氯仿溶解,制成标准对照品储备液。另精密称取二十三烷5.7mg,置1ml量瓶中,加氯仿至刻度,为内标储备液。
(3)校正因子测定 用100μl微量注射器吸取标准对照品储备液160μl置于1ml量瓶中,再于量瓶中加入100μl内标储备液,加氯仿至刻度,混匀,取1μl注入气相色谱仪,记录色谱图,用峰面积计算校正因子。
(4)样品溶液的制备 取土木香药材粉末约1g,精密称定,置具塞瓶中,准确加入氯仿3ml,冷浸过夜,取滤液适量,加内标溶液120μl,氯仿定容至1ml,作为样品溶液。
(5)测定 吸取样品溶液1μl,注入气相色谱仪,按内标法计算药材中土木香内酯和异土木香内酯的百分含量。

【对多发性骨髓瘤 RPMI-8226 细胞增殖、凋亡的影响】[2]
使用浓度为 1、2. 5、5、7. 5 及 10 μmol/L 的土木香内酯处理 RPMI-8226 细胞 48 h,应用 CCK-8检测细胞活力,并计算其半数抑制浓度( IC50) 值; 使用浓度为 2. 5,5 及 7. 5 μmol/L 的土木香内酯处理 RPMI-8226细胞 48 h 后,应用 Annexin V/PI 双标记法分析细胞凋亡; Western blot 方法检测呈切割的 caspase-3 及 ERK 信号通路的变化; 裸鼠荷瘤并给予土木香内酯以进一步验证其对多发性骨髓瘤细胞的促凋亡作用。发现土木香内酯可抑制 RPMI-8226 细胞活力,并呈现剂量依赖效应( r = - 0. 9784,P = 0. 0007) ; RPMI-8226 细胞 48 h 的 IC50为 4. 32 ±0. 15 μmol / L; 流式细胞术分析凋亡结果显示,随着土木香内酯浓度的增高,细胞早期凋亡率显著增加( r = 0. 9601,P< 0. 05) ; Western blot 结果显示,土木香内酯处理 RPMI-8226 细胞后,活化了 caspase-3,降低 ERK 磷酸化水平; 体内结果显示,给药组小鼠瘤体积明显小于对照组( P < 0. 05) ; 免疫组织化学检测结果显示,与对照组相比,实验组瘤内细胞核增殖相关抗原 Ki67 及 p-ERK 的水平降低。结论: 土木香内酯可有效地抑制多发性骨髓瘤 RPMI-8226 细胞增殖并促进其凋亡,其机制可能是通过抑制 ERK 信号通路的活性,抑制裸鼠皮下多发性骨髓瘤的生长。
已有研究发现,土木香内酯具有多种抗肿瘤的效果。本研究选择多发性骨髓瘤 RPMI-8226 细胞作为研究对象,通过体外和体内两方面研究土木香内酯对多发性骨髓瘤的抗肿瘤作用。首先应用CCK-8 检测土木香内酯对 RPMI-8226 细胞的杀伤作用,发现其可以显著的抑制骨髓瘤的细胞活性,48 hIC50为 4. 32 ± 0. 15 μmol/L。此前,Mi 等报道人正常干细胞株 L02 对土木香内酯较不敏感,其 IC50约为 31. 34 μg /ml( 133.75 μmol/L) 。由此可见,土木香内酯对正常细胞杀伤力较低,而在较低的浓度下对骨髓瘤细胞杀伤效果明显。进一步的流式细胞术检测土木香内酯处理骨髓瘤细胞后发现,土木香内酯通过诱导细胞凋亡发挥抗骨髓瘤的作用。
