The general procedure for the synthesis of 4-iodo-2-aminopyridine from 2-fluoro-4-iodopyridine was as follows: 2-fluoro-4-iodopyridine (10.00 g, 43.50 mmol) was mixed with ammonium hydroxide (10 mL) in DMSO (20 mL), and the reaction was stirred for 40 h at 100 °C. After completion of the reaction, water (100 mL) was added to the reaction mixture and the precipitate was collected by filtration to afford the target product 4-iodo-2-aminopyridine (Compound 10a) as a brown solid in 90% yield (8.62 g).
