Preparation of methyl 5-(bromomethyl)-2-fluorobenzoate (3): methyl 2-fluoro-5-(hydroxymethyl)benzoate (2) (5 g, 27.17 mmol, 1 eq.) was dissolved in anhydrous dichloromethane (50 mL) followed by addition of phosphorus tribromide (2.19 g, 8.12 mmol, 0.3 eq.). The reaction mixture was stirred at room temperature for 3 hours. Upon completion of the reaction, the reaction was quenched with saturated sodium bicarbonate solution and extracted with dichloromethane (3 x 100 mL). The organic phases were combined, washed sequentially with water (3 x 30 mL) and brine (1 x 30 mL), dried over anhydrous sodium sulfate, filtered and concentrated to afford methyl 5-(bromomethyl)-2-fluorobenzoate (3) (4.8 g, 71% yield) as an off-white solid. | [References]
