Tert-butyl 6,7-dihydrothieno[3,2-c]pyridine-5(4H)-carboxylate (5.0 g, 20.89 mmol) was used as starting material and dissolved in acetonitrile (25 mL). To this solution was added N-bromosuccinimide (4.09 g, 22.98 mmol) at room temperature and the reaction mixture was stirred overnight. Upon completion of the reaction, the reaction was quenched by the addition of water (50 mL) and subsequently extracted with ethyl acetate (50 mL × 2). The organic phases were combined and washed sequentially with water (50 mL × 2) and saturated brine (40 mL), then dried over anhydrous sodium sulfate. The solvent was removed by concentration under reduced pressure, and the crude product obtained was purified by column chromatography (eluent: petroleum ether/ethyl acetate, v/v = 20/1) to give the final white solid product tert-butyl 2-bromo-6,7-dihydrothieno[3,2-c]pyridine-5(4H)-carboxylate (6.21 g, 93% yield). | [References]
