General procedure for the synthesis of 6-N-morpholinopyrimidin-4-amine from 4-amino-6-chloropyrimidine and morpholine: 400 mg of 4-amino-6-chloropyrimidine and 1.51 g (1.5 eq.) of cesium carbonate were added to a reaction flask, 10 mL of N,N-dimethylformamide (DMF) was added as solvent followed by 405 μL (1.5 eq.) of morpholine. The reaction mixture was heated to 100 °C and stirred for 12 hours. After completion of the reaction, an appropriate amount of water was added and the organic phase was extracted with ethyl acetate. The organic phase was washed with saturated brine, dried over anhydrous sodium sulfate, filtered and the solvent was evaporated under reduced pressure. The residue was separated and purified by silica gel column chromatography using methanol/dichloromethane (1:50, v/v) as eluent, resulting in 245 mg of yellow solid product in 44% yield.
