向氢化钠(2.77 g,69.2 mmol)的无水乙醇(150 mL)悬浮液中分批加入2,4-二氨基-6-氯嘧啶(5.0 g,34.6 mmol)。将反应混合物加热至回流并保持6小时。反应完成后,冷却至室温,用5-6 N盐酸的异丙醇溶液中和反应液。减压浓缩除去溶剂,所得残余物通过硅胶柱色谱法(洗脱剂:二氯甲烷/甲醇=40:1,v/v)进行纯化,得到2,4-二氨基-6-乙氧基嘧啶,为白色固体(4.0 g,收率75%)。产物熔点为164-165℃。1H NMR(300 MHz,DMSO-d6,25℃):δ 6.00(s,2H,NH2),5.88(s,2H,NH2),5.03(s,1H,CH),4.13(q,J=7.1 Hz,2H,OCH2CH3),1.22(t,J=7.1 Hz,3H,OCH2CH3)。13C NMR(75 MHz,DMSO-d6,25℃):δ 170.0(C),165.9(C),162.9(C),76.1(CH),60.2(OCH2CH3),14.7(OCH2CH3)。高分辨质谱(HRMS):计算值C6H11N4O [M+H]+ 155.09329,实测值155.09260。
