在500 mL圆底烧瓶中,将4-(6-硝基吡啶-3-基)-5,6-二氢吡啶-1(2H)-羧酸叔丁酯(4.9 g,16.0 mmol,1.00当量)溶于乙醇(300 mL)和乙酸乙酯(75 mL)的混合溶剂中,加入钯碳(1.32 g,1.24 mmol,0.0773当量)。将反应混合物用氢气置换两次,然后在氢气氛围下(使用氢气气球)搅拌过夜。通过LC/MS分析确认反应完成。反应混合物用氮气吹扫后,通过硅藻土垫过滤,硅藻土垫用乙酸乙酯洗涤数次。合并无色滤液和洗涤液,加入二氯甲烷,将溶液蒸发至干。再次加入二氯甲烷,溶液经真空浓缩,得到目标产物4-(6-氨基吡啶-3-基)哌啶-1-甲酸叔丁酯,收率定量。(M + H)+ = 278 m/z。
