以7-甲氧基-2-萘甲腈为原料合成7-羟基-2-萘甲腈的一般步骤:在氮气保护下,将三溴化硼(840μL,8.8mmol)的二氯甲烷(4mL)溶液缓慢滴加至7-甲氧基-2-萘甲腈(646mg,3.53mmol)的二氯甲烷(6mL)溶液中。反应混合物在室温下搅拌30小时。反应完成后,缓慢加入去离子水(约5mL)淬灭反应。用饱和碳酸钠溶液调节反应混合物的pH至7,随后用二氯甲烷萃取。合并有机层,用无水硫酸钠干燥,过滤后减压浓缩除去溶剂。粗产物通过硅胶柱色谱法纯化,洗脱剂为二氯甲烷与甲醇的混合溶液(梯度洗脱:1%甲醇的二氯甲烷溶液至5%甲醇的二氯甲烷溶液),得到7-羟基-2-萘甲腈,为白色固体(477mg,收率80%)。高分辨质谱(EI)分析结果:C11H7NO的计算值为169.0528,实测值为169.0527。
