以5-甲基-4,5,6,7-四氢噻唑并[5,4-c]吡啶-2-胺(12)(600.0g,3.545mol)为原料,将其悬浮于水(6.0L)中。在0至15℃下,缓慢滴加48%氢溴酸(4.2L)。随后,在1.5至5小时内滴加亚硝酸钠(367.2g,3.56mol)的水(1.8L)溶液。滴加完毕后,将反应混合物加热至30℃并持续搅拌24小时。反应完成后,用5N氢氧化钠水溶液(6.0L)中和反应混合物至强碱性(pH = 12.5)。然后,用甲苯(12.0L和6.0L)分两次萃取水层。合并甲苯层,加入无水硫酸钠(1202.0g)干燥。过滤除去不溶物后,将母液在40℃下减压浓缩,得到目标产物2-溴-5-甲基-4,5,6,7-四氢噻唑并[5,4-c]吡啶(11a)(557.6g,收率67.5%)。产物经1H-NMR(CDCl3)表征:δppm 3.58-3.57(t,3H,J = 1.8Hz),2.92-2.87(m,2H),2.81-2.76(m,2H),2.49(s,3H)。
