以4-溴-5-氟-2-硝基苯胺为原料合成4-溴-5-氟苯-1,2-二胺的一般步骤:向4-溴-5-氟-2-硝基苯胺(1.0g,4.3mmol)的THF(9.0mL)溶液中加入EtOH(9.0mL)和H2O(3mL),随后加入铁粉(1.2g,21.3mmol)和氯化铵(0.34g,6.4mmol)。将反应混合物在95℃下加热回流4小时。反应完成后,将混合物冷却至室温,用EtOH稀释,通过硅藻土垫过滤,直至滤液无色。滤液经减压浓缩后,将残余物溶解于EtOAc中,依次用H2O和盐水洗涤,有机相用无水Na2SO4干燥,过滤并浓缩。向浓缩物中加入己烷,过滤收集析出的固体,得到标题化合物4-溴-5-氟苯-1,2-二胺(710mg,3.5mmol,收率81%)。
