通用方法：在0℃下，将D-天冬氨酸-4-叔丁酯（1.89g，10mmol）溶于四氢呋喃（20mL）和水（12mL）的混合溶剂中。搅拌5分钟后，分次缓慢加入二碳酸二叔丁酯（4.35g，20mmol），随后加入碳酸钠（4.20g，40mmol）。将反应混合物在室温下搅拌24小时，反应完成后，于减压条件下浓缩去除溶剂。将残余物溶解于10%碳酸氢钠水溶液（50mL）中，用乙醚（3×20mL）洗涤水相。用柠檬酸调节水相pH至4.0后，用乙酸乙酯（3×100mL）萃取产物。合并有机相，用无水硫酸镁干燥，过滤后减压浓缩，得到目标产物Boc-O-叔丁酯-D-天冬氨酸（4-叔丁氧基-2-（叔丁氧基羰基氨基）-4-氧代丁酸），为无色油状物（2.82g，收率97%）。
