一般步骤:向化合物11(500 mg,0.87 mmol)和双(4-硝基苯基)碳酸酯(双-PNP,2.64 g,8.72 mmol)的DCM:DMF(8:2,25 mL)混合溶液中加入N,N-二异丙基乙胺(DIPEA,0.45 mL,2.61 mmol)。反应混合物在23℃下搅拌20小时后,通过硅胶柱色谱(洗脱剂:DCM:CH3OH,50:1至10:1)纯化,得到目标产物马来酰亚胺基己酰-L-缬氨酸-L-瓜氨酸对氨基苄醇对硝基苯基碳酸酯9(364 mg,收率57%)。薄层色谱(TLC)Rf = 0.40(展开剂:CH2Cl2:CH3OH,9:1)。1H NMR(400 MHz,CDCl3/CD3OD)δ 9.45(s,1H),8.23(d,J = 8.3 Hz,2H),7.59(d,J = 8.5 Hz,2H),7.35(d,J = 8.3 Hz,2H),7.34(d,J = 8.5 Hz,2H),6.65(s,2H),5.20(s,2H),4.56(dt,J = 10.5, 5.4 Hz,1H),4.15(d,J = 7.2 Hz,1H),3.46(dd,J = 8.0, 6.4 Hz,2H),3.16-2.89(m,2H),2.21(dd,J = 8.3, 6.6 Hz,2H),2.06-1.97(m,1H),1.90-1.83(m,1H),1.73-1.46(m,7H),1.34-1.20(m,2H),0.91(d,J = 6.7 Hz,3H),0.90(d,J = 6.7 Hz,3H)。13C NMR(125 MHz,CDCl3/CD3OD)δ 174.4, 172.4, 171.1, 170.6, 160.5, 155.5, 152.5, 145.3, 138.7, 134.1, 129.9, 129.5, 125.2, 121.8, 120.0, 70.6, 59.0, 53.2, 37.5, 35.8, 30.6, 29.6, 29.3, 28.1, 26.2, 25.1, 19.1, 18.1。ESI-MS m/z:计算值C34H42N6O11:737.3;实测值:738.3([M + H]+)。
