以5-烯丙基-4,6-二氯嘧啶为原料合成4,6-二氯嘧啶-5-乙醛的一般步骤如下:在配备磁力搅拌器的50 mL三颈烧瓶中,依次加入20 mL二氯甲烷和4,6-二氯-5-烯丙基嘧啶(5.0 g,0.026 mol),随后加入二甲基亚砜(8 g,0.104 mol)和三乙胺。将反应混合物冷却至-40°C后,通入臭氧(O3)直至反应溶液呈现蓝色,此时停止臭氧通入并切换为氮气(N2)保护,持续30分钟以彻底排除体系中的臭氧。接着,向反应体系中加入硫脲(2.0 g,0.026 mol),并允许反应混合物自然升温至室温。使用淀粉碘化钾试纸检测反应体系,确认无过氧化物存在(试纸不变蓝)。最后,向反应混合物中加入60 mL水,并用二氯甲烷进行两次萃取。该步骤的收率为88%。
