步骤1:中间体3-甲基噻唑烷-2,4-二酮(33a)的制备 将噻唑烷-2,4-二酮(2g,17.1mmol)、碳酸钾(3.55g,27.7mmol)和碘甲烷(1.87mL,30mmol)溶于N,N-二甲基甲酰胺(16mL)中,于75℃搅拌反应3小时。反应完成后,将混合物过滤并浓缩。残余物用水(30mL)和乙酸乙酯(30mL)稀释,分离有机层。有机层依次用水(2×10mL)和盐水(30mL)洗涤,无水硫酸钠干燥,过滤后减压浓缩。经硅胶柱层析(洗脱剂:二氯甲烷)纯化,得到目标产物3-甲基噻唑烷-2,4-二酮(33a),为白色固体(1.77g,13.5mmol,收率79%)。 1H NMR (400 MHz, CDCl3) δ: 3.19 (s, 3H), 3.95 (s, 2H). MS m/z ([M+H]+) 132.
