以2,4-二氯吡啶和甲醇钠为原料合成2,4-二甲氧基吡啶的一般步骤:将新制备的甲醇钠(24 g,0.44 mol)加入到2,4-二氯吡啶(15 g,0.10 mol)的无水N-甲基-2-吡咯烷酮(80 mL)溶液中。将反应混合物在120℃下搅拌6小时。反应完成后,将混合物冷却至室温,用乙酸乙酯(800 mL)稀释,并用水洗涤。分离有机层,用无水硫酸钠干燥,过滤后浓缩,得到2,4-二甲氧基吡啶(4),为无色油状物,其纯度足以用于下一步骤。产量:11.2 g(80%)。1H NMR (400 MHz, DMSO-d6): δ 3.80 (s, 3H), 3.84 (s, 3H), 6.33 (d, J = 2.0 Hz, 1H), 6.60 (dd, J = 2.0 Hz, J = 5.6 Hz, 1H), 7.97 (d, J = 5.6 Hz, 1H); 13C NMR (100 MHz, DMSO-d6): δ 167.5, 165.4, 147.4, 106.1, 93.8, 55.3, 53.1; HRMS (ESI) m/z calcd for C7H10NO2 [M+H]+: 140.0706; found: 140.0707.
