以2-氰基吡嗪为原料合成2-氨甲基吡嗪的一般步骤如下:参照文献(JP A 2001-894594)所述方法,具体操作如下。原料2-氰基吡啶购自Sigma-Aldrich Co.LLC。将1.05g(10mmol)的2-氰基吡啶和100mg的60wt% Ni/SiO2催化剂置于20mL甲苯中,装入高压釜(型号5U5316)内,随后用氩气置换釜内空气。在氢气氛围下加压至50atm,于140℃反应温度下搅拌反应4小时。反应完成后,将反应液过滤除去催化剂,滤液经浓缩处理,定量得到目标产物2-氨甲基吡啶(2-AMPZ)。产物经1H-NMR(399.78MHz, CDCl3)表征:δ 8.60-8.45(m, 3H), 4.07(s, 2H), 1.79(br, 2H)。
