在0℃条件下，将N-碘代丁二酰亚胺（NIS，2.25g，10.0mmol）分批加入至2,6-二氟苯甲酸（1.58g，10.0mmol）的浓硫酸（25mL）溶液中。反应混合物在0℃下持续搅拌3.5小时。随后，将混合物缓慢倒入碎冰（10g）中，并加入3%的亚硫酸钠溶液（10mL）以淬灭反应。所得浆液在0℃下继续搅拌1小时，之后通过过滤分离产物。产物用水洗涤并干燥，得到目标化合物2,6-二氟-3-碘苯甲酸（化合物3），为白色固体（2.05g，收率72%）。产物结构通过1H NMR（CDCl3）δ6.78-6.82（t，1H，J=8.4Hz，ArH），7.78-7.83（m，1H，ArH）和GC-MS（m/z：240 [M-44]+）进行确认。
