在0℃下,向搅拌中的1-叔丁氧羰基哌啶-4-甲醛(358 mg,1.68 mmol)和碳酸钾(464 mg,3.36 mmol)在甲醇(16 mL)中的混合物中,缓慢滴加(1-重氮基-2-氧代丙基)膦酸二甲酯(323 mg,1.68 mmol)的甲醇(2 mL)溶液。将反应混合物在室温下搅拌过夜,随后过滤并浓缩。残余物通过硅胶柱色谱纯化,使用乙酸乙酯/己烷(1:5,v/v)作为洗脱剂,得到1-Boc-4-炔基哌啶(308 mg,收率88%)为无色晶体。LC-MS m/z: 154 [M-Boc+2H]+。(参考文献:J. Am. Chem. Soc. 2003, 125, 3714。)
