步骤3:3-叔丁基3-甲基(S)-哌嗪二甲酸酯(AA3)的合成;在室温条件下,向搅拌中的AA 2的甲醇溶液(0.1M)中加入10%Pd-C(0.2当量)。将反应混合物置于氢气气氛下,室温搅拌3小时。反应完成后,过滤混合物,并用甲醇洗涤滤饼。将滤液减压浓缩,得到目标产物(S)-1-N-Boc-3-哌嗪甲酸甲酯,收率为89%。产物经1H NMR(400MHz,CDCl3,300K)确认:δ4.05(1H,m),3.76(3H,s),3.74-3.68(1H,m),3.44(1H,dd,J = 8.6,3.5Hz),3.30-3.12(1H,m),3.10-3.00(2H,m),2.75(1H,m),2.17-1.74(1H,m),1.47(9H,s)。质谱分析(ES +)显示:C11H20N2O4的理论分子量为244,实测值为267(M + Na)+。
