向0℃下搅拌的中间体1（8.2g，30mmol）和三乙胺（6.07g，8.36mL，60mmol）在无水二氯甲烷（120mL）中的溶液中缓慢加入甲磺酰氯（5.49g，3.71mL，48mmol）。在0℃下反应2小时后，额外加入三乙胺（6mmol）和甲磺酰氯（4.8mmol）。反应2小时后，薄层色谱（TLC，展开剂：己烷/乙酸乙酯，1:1）显示反应完全。将反应混合物用二氯甲烷（120mL）稀释，依次用饱和碳酸氢钠溶液（2×240mL）和水（2×240mL）洗涤，有机相经干燥、过滤并浓缩，得到5-(氯甲基)-4-甲基-2-[4-(三氟甲基)苯基]-1,3-噻唑（8.0g，27mmol，产率90%）为黄色固体。
