步骤B. 在0℃下,将4-氟-3-硝基苯胺(5.0g,32.0mmol)和三乙胺(TEA,6.7mL,48.0mmol)溶解于50mL二氯甲烷(DCM)中。随后,缓慢滴加乙酰氯(2.75mL,38.4mmol),滴加完毕后将反应混合物升温至室温并搅拌过夜。反应完成后,依次用5%硫酸氢钾(KHSO4)水溶液、饱和碳酸氢钠(NaHCO3)水溶液和盐水洗涤有机层,然后用无水硫酸镁(MgSO4)干燥。通过浓缩有机相并从DCM中结晶,得到N-(4-氟-3-硝基苯基)乙酰胺5.3g,产率84%。产物结构经1H NMR(400MHz,氯仿-D)确认:δ2.04(s,3H),7.51(dd,J = 11.23,9.08Hz,1H),7.80(ddd,J = 9.08,4.00,2.93Hz,1H),8.47(dd,J = 7.03,2.73Hz,1H),10.38(s,1H)。
