以2-溴苯胺(2.5g,14.62mmol)和乙氧基亚甲基丙二酸二乙酯(3.16g,14.62mmol)为原料,在100℃下加热反应3小时。随后,通过氮气流除去反应体系中的挥发物。将所得熔融物缓慢加入沸腾的二苯醚(10mL)中,并将混合物加热回流2小时。反应完成后,待反应混合物冷却至室温,加入石油醚以沉淀产物。通过过滤收集沉淀的固体,干燥后得到4-羟基-8-溴喹啉-3-羧酸乙酯(3.5g)。产物结构经1H NMR(300MHz,DMSO-d6)确认:δ11.65(br s,1H),8.45(s,1H),8.17(d,J = 7.8Hz,1H),8.05(d,J = 7.8Hz,1H),7.39-7.34(t,J = 7.8Hz,1H),4.26-4.19(q,J = 7.2,14.1Hz,2H),1.30-1.26(t,J = 6.9Hz,3H)。
