以2-氯-4-氰基吡啶为原料合成4-氰基-2-氟吡啶的一般步骤:将2-氯-4-氰基吡啶(6.0 g,43.5 mmol)、氟化钾(7.56 g,130.3 mmol)和溴化四丁基鏻(14.8 g,43.7 mmol)溶于1-甲基-2-吡咯烷酮(20 mL)中,加热至100°C反应18小时。反应完成后,用水稀释反应混合物,并用乙酸乙酯(EtOAc)萃取。有机相依次用水和盐水洗涤,无水硫酸钠(Na2SO4)干燥,减压浓缩,得到目标产物4-氰基-2-氟吡啶(2.3 g,收率43%)。质谱(ES)显示m/z 123.1([M + H]+)。核磁共振氢谱(400 MHz,CDCl3)δ 8.43(d,1H,J = 5.2 Hz),7.45(m,1H),7.22(m,1H)。
