一般步骤:将37%甲醛水溶液(1.26 mL,16.9 mmol)缓慢加入至(2R)-哌啶-2-羧酸(2.00 g,15.5 mmol)与5% Pd/C(500 mg)的甲醇(20 mL)悬浮液中。反应体系经氢气置换后,补加甲醛(0.60 mL,8.06 mmol),在氢气氛围下室温搅拌20小时。反应完成后,混合物经硅藻土垫抽滤,甲醇(200 mL)洗涤滤饼。合并滤液减压浓缩,得到白色固体状标题化合物(R)-1-甲基-2-哌啶甲酸(2.23 g,定量收率),直接用于后续反应。TLC检测:Rf = 0.25(展开剂V(CH2Cl2)/V(MeOH)=1:1)。熔点:208-210℃。比旋光度:[α]D27.1 = +67.6(c 1.27,甲醇)。红外光谱(KBr压片):3407, 2950, 1615, 1399 cm-1。1H NMR(400 MHz, CDCl3):δ 8.22(宽峰,1H),3.64(d,1H,J = 12.0 Hz),3.28(多重峰,1H),2.86(s,3H),2.70(多重峰,1H),2.34(d,1H,J = 14.4 Hz),2.00-1.74(多重峰,4H),1.46(多重峰,1H)。13C NMR(100 MHz, DMSO-d6):δ 170.4, 68.3, 53.3, 42.3, 28.0, 23.1, 21.8。高分辨质谱(ESI):m/z计算值C7H13NO2 [M]+ 143.0946,实测值143.0947。
