以2,7-二溴咔唑和碘苯为原料合成2,7-二溴-9-苯基-9H-咔唑的一般步骤:将32.5g(100毫摩尔)2,7-二溴-9H-咔唑、20.4g(100毫摩尔)碘苯、9.5g(150毫摩尔)铜粉、27.6g(200毫摩尔)碳酸钾和600ml二甲基甲酰胺混合,在氮气保护下于130℃加热反应过夜。反应完成后,冷却至室温,过滤反应混合物。滤液用二氯甲烷和水进行三次萃取,有机相用无水硫酸镁干燥,随后在真空下蒸发除去溶剂。残余物通过硅胶柱色谱法(洗脱剂:己烷-二氯甲烷)纯化,得到白色固体产物2,7-二溴-9-苯基-9H-咔唑(31.3g,78毫摩尔,收率78%)。
