步骤1:4-氟-3-碘-1H-吲唑(B-2)的制备。在室温下,向搅拌中的4-氟-1H-吲唑(B-i)(5.00 g,36.73 mmol)的DMF(80 mL)溶液中依次加入碘(I2)(18.64 g,73.46 mmol)和氢氧化钾(KOH)(7.73 g,137.7 mmol)。反应2小时后,薄层色谱(TLC)监测显示反应完成。将反应混合物缓慢倒入10%的亚硫酸氢钠(NaHSO3)水溶液(200 mL)中,并用乙酸乙酯(EA)(3×200 mL)萃取。合并有机层,依次用水(100 mL)和饱和食盐水(2×200 mL)洗涤,经无水硫酸钠(Na2SO4)干燥,过滤后浓缩。粗产物用石油醚(PE)洗涤,得到黄色固体B-2(8.33 g),产率为86.5%。B-2的物理表征数据如下:液相色谱-质谱联用(LCMS)(电喷雾离子化,ESI):计算值C7H4FIN2,261.9;实测值[M + H]+=262.9。
