以1H-咪唑-4-甲酸为原料合成咪唑并[1,5-a:1',5'-d]吡嗪-5,10-二酮的一般步骤:将1H-咪唑-4-甲酸(5g,44.6mmol)悬浮于亚硫酰氯(75mL)中,搅拌下加热回流18小时。反应完成后,冷却反应混合物,过滤,用甲苯洗涤,并在高真空下干燥,得到目标化合物咪唑并[1,5-a:1',5'-d]吡嗪-5,10-二酮,为黄色固体,收率95%。分子式:C8H4N4O2,分子量:188.14g/mol,熔点:265-268℃。1H NMR (300 MHz, DMSO-d6) δ: 8.24 (d, 1H, CHC-CO), 8.32 (d, 1H, CHC-CO, J=3Hz), 8.88 (d, 1H, CHN), 9.28 (d, 1H, CHN, J=3Hz)。13C NMR (100 MHz, DMSO-d6) δ: 123.99, 124.52, 125.62, 137.26, 139.73, 150.39, 159.68。MS (ESI+, QTof, m/z): 189.0 [M + H]+。
