以对氨基苯乙酸(20g,0.132mol)和甘油(51g,0.555mol)为原料,合成6-喹啉乙酸的一般步骤如下:将氨基苯乙酸、FeSO4(4.9g,0.032mol)、甘油、PhNO2(10.5g,0.086mol)和浓H2SO4(23mL)混合,加热至200℃并维持5小时。反应完成后,用4N NaOH水溶液调节反应混合物的pH至10。过滤反应混合物,将澄清的滤液用AcOH酸化至pH约5。过滤收集生成的沉淀,得到6-喹啉乙酸(12.5g,产率51%),为棕色固体。TLC条件:20% MeOH/CH2Cl2,Rf=0.2。1H NMR(300MHz,DMSO-d6)δ ppm:8.85(m,1H),8.31-8.28(m,1H),7.96(d,J=8.3Hz,1H),7.82(m,1H),7.67-7.64(m,1H),7.50-7.49(m,1H),3.78(s,2H)。
