在0℃条件下,将3-羟基-2-吡啶甲酸(10g,72mmol,1当量)的甲醇(150mL,0.5M)悬浮液缓慢滴加至浓硫酸(12mL,216mmol,3当量)中。反应混合物在回流条件下搅拌6小时。反应完成后,将混合物冷却至室温,并在减压下浓缩。随后,用饱和NaHCO3水溶液和固体NaHCO3调节pH至8.5。水层用EtOAc萃取,合并的有机层用盐水洗涤,经MgSO4干燥,减压浓缩,得到3-羟基-2-吡啶甲酸甲酯,为白色固体(10.9g,99%)。薄层色谱(TLC)显示Rf = 0.3(石油醚/EtOAc 1:1,v/v)。熔点为74°C。1H NMR(300MHz,CDCl3):δ(ppm)= 4.06(s,3H),7.34(dd,J = 3.9,8.4Hz,1H),7.38(dd,J = 1.5,8.4Hz,1H),8.38(dd,J = 1.5,3.9Hz,1H),10.64(s,1H)。13C NMR(75MHz,CDCl3):δ= 53.2,126.3,129.8,130.2,141.6,158.9,169.9。质谱(ESI+):m/z(百分比):154(100)[M + H]+。
