以3-(二甲基氨基)-1-(吡啶-3-基)丙-2-烯-1-酮(15.4g,0.088mol)、硝酸胍(10.7g,0.088mol)和氢氧化钠(3.5g,0.088mol)为原料,溶于正丁醇(120mL)中。在搅拌下将混合物加热至回流状态,并维持反应16小时。反应完成后,将混合物冷却至室温。通过过滤收集固体产物,并用去离子水(400mL)洗涤。将所得4-(3-吡啶基)-2-氨基嘧啶在真空条件下干燥3天,得到12.7g黄色晶体产物,产率为85%。产物结构经1H NMR(500MHz,CDCl3)确认:δ5.15(宽峰,2H),7.09(双峰,1H),7.44(多重峰,1H),8.33(双峰,1H),8.42(双峰,1H),8.73(双峰,1H),9.22(单峰,1H)。LC-MS分析显示:m/z计算值为172.2,实测值为173.2 [M + H]+。
