通用方法:将五氧化二磷(2.02 g,14.23 mmol)溶解于甲磺酸(13 mL,195.8 mmol)中,加热至80℃并搅拌至均匀。随后,加入3-[(1H-吡咯-2-羰基)-氨基]丙酸(3.24 g,17.78 mmol),持续搅拌反应并通过薄层色谱(TLC)监测反应进程。反应完成后,用冰水淬灭反应混合物,并用乙酸乙酯进行多次萃取。合并有机相,用无水硫酸镁(MgSO4)干燥,减压浓缩得到6,7-二氢吡咯并[2,3-c]氮杂卓-4,8(1H,5H)-二酮,为白色固体(1.58 g,产率54%)。产物表征数据如下:熔点280-282℃;IR (cm-1) 3222, 2931, 1659, 1634;1H NMR (δ, ppm) 2.70 (2H, m), 3.35 (2H, m), 6.55 (1H, d, J = 2.7 Hz), 6.99 (1H, d, J = 2.7 Hz), 8.34 (1H, t, J = 4.7 Hz), 12.18 (1H, br s);13C NMR (δ, ppm) 36.9, 43.8, 109.9, 122.7, 123.9, 128.3, 162.6, 194.7;LRMS (ES+) m/z 186.8 [M + Na]+;HRMS (ES+) 165.0657 ([M + H]+, 计算值C8H9N2O2为165.0664)。
