通用方法:将5-溴-2-硝基吡啶(24.7 mmol)、哌嗪(29.2 mmol)和N,N-二异丙基乙胺(DIPEA,37.1 mmol)在乙腈(100 mL)中的混合物回流8小时。通过薄层色谱(TLC)监测反应进程,待原料完全转化后,将反应混合物冷却至室温。蒸发去除溶剂,残余物通过柱色谱法纯化,得到1-(6-硝基吡啶-3-基)哌嗪(8c)。产物为黄色固体,收率82%。1H NMR(DMSO-d6, 400 MHz)δ:7.96(d, J = 9.3 Hz, 1H, Pyr-H),7.88(d, J = 2.9 Hz, 1H, Pyr-H),7.26(dd, J = 9.3, 3.0 Hz, 1H, Pyr-H),3.64(t, 4H, Pyr-NCH2CH2N),3.32(t, 4H, Pyr-NCH2CH2N)。13C NMR(DMSO-d6, 100 MHz):δ 151.7, 146.2, 136.6, 125.9, 118.4, 51.3, 45.8。高分辨质谱(HRMS, ESI):C9H12N4O2的计算值[M + H]+为209.1039,实测值为209.1030。
