步骤1. 在2L三颈烧瓶中依次加入4-氯-2-(甲硫基)嘧啶-5-羧酸乙酯(100g, 0.43mol)、四氢呋喃(THF, 500mL)和三乙胺(TEA, 186mL, 1.29mol)。在搅拌下分批加入28%氢氧化铵水溶液(400mL),控制反应温度低于30℃。反应2小时后,加入水(1000mL),通过真空蒸馏除去THF。将反应混合物过滤,收集固体产物,并在50℃下真空干燥,得到白色固体4-氨基-2-(甲硫基)嘧啶-5-羧酸乙酯(90g, 收率99%)。产物经LC/MS分析显示m/z = 213.9 [M+H]+;1H-NMR (DMSO-d6, 400MHz) δ 8.57 (s, 1H), 8.03 (br, 1H), 7.65 (br, 1H), 4.27 (q, J = 7.5Hz, 2H), 2.46 (s, 3H), 1.29 (t, J = 7.2Hz, 3H)。
