以4-吡唑甲酸乙酯和碘甲烷为原料合成1-甲基-1H-吡唑-4-羧酸乙酯的一般步骤:将1H-吡唑-4-羧酸乙酯(2.5 g,17.9 mmol)和碘甲烷(3.1 g,21.5 mmol)溶解于N,N-二甲基甲酰胺(DMF,10 mL)中,随后加入碳酸钾(K2CO3,5.0 g,35.8 mmol)。将反应混合物在室温下搅拌16小时。反应完成后,向混合物中加入水(50 mL),然后用乙酸乙酯(EA,50 mL ×2)进行萃取。合并有机层,用饱和食盐水(40 mL)洗涤,无水硫酸钠(Na2SO4)干燥,减压浓缩。通过硅胶柱色谱法(洗脱剂:石油醚/乙酸乙酯 = 8:1)纯化残余物,得到1-甲基-1H-吡唑-4-羧酸乙酯(214 mg,收率:85%),为无色油状物。1H NMR(300 MHz,DMSO-d6):δ = 8.28(s,1H),7.82(s,1H),4.26(q,J = 6.9 Hz,2H),3.87(s,3H),1.25(t,J = 6.9 Hz,3H)。质谱(MS):m/z 155.3([M + H]+)。
