向搅拌的甲醛水溶液(37% w/w,37.5 mmol,2.8 mL)和二甲胺水溶液(40% w/w,37.5 mmol,4.8 mL)的乙醇(25 mL)溶液中加入4-羟基-3-甲氧基苯甲醛(香兰素,25 mmol,3.883 g)。将反应混合物搅拌并加热回流45分钟,随后在室温下继续搅拌过夜。反应完成后,将析出的乳白色固体通过减压过滤分离,用冰冷的丙酮洗涤,并在室温下真空干燥,得到3-((二甲氨基)甲基)-4-羟基-5-甲氧基苯甲醛,产物为灰白色固体。产率:75%(3.910 g);熔点:140-142°C(文献值:139-141°C,参见参考文献[43j]);1H NMR(400 MHz,CDCl3)δ(ppm):10.6(1H,宽单峰,OH),9.77(1H,单峰,CHO),7.34(1H,单峰,ArH),7.15(1H,单峰,ArH),3.94(3H,单峰,OCH3),3.76(2H,单峰,CH2),2.38(6H,单峰,2×CH3);13C NMR(100 MHz,CDCl3)δ(ppm):190.7,154.5,148.6,128.1,125.3,121.4,109.8,62.2,56.0,44.3。
