(a)(3S)-4-苄基-3-(氯甲基)吗啉的合成:在氩气保护下,将[(3R)-4-苄基吗啉-3-基]甲醇(1.83 g,8.8 mmol)溶解于无水二氯甲烷(15 mL)中。向该溶液中依次加入亚硫酰氯(3.15 g,26.5 mmol)和N,N-二甲基甲酰胺(DMF,2滴)。将反应混合物加热至回流,持续搅拌2小时30分钟。反应完成后,通过旋转蒸发仪除去溶剂。将残余物用饱和碳酸氢钠水溶液中和,随后用乙酸乙酯(3 × 15 mL)萃取。合并有机相,用无水硫酸钠干燥,过滤后浓缩,得到油状产物(3S)-4-苄基-3-(氯甲基)吗啉(1.88 g,收率94%)。产物通过1H NMR(500 MHz,CDCl3)和LCMS进行表征:1H NMR (500 MHz, CDCl3) δ 2.3-2.4 (m, 1H), 2.7 (m, 1H), 2.8 (m, 1H), 3.5 (d, 1H), 3.6-3.9 (m, 5H), 4.0 (d, 1H), 7.3 (m, 1H), 7.4 (m, 4H); LCMS: m/z 226 [M + H]+。
