向2,4,6-三氯嘧啶-5-甲醛(8g,37.84mmol,1.00当量)的乙醇(120mL)溶液中依次加入40%甲基肼水溶液(4mL,37.98mmol,1.00当量)和三乙胺(16mL)。反应混合物在-78℃下搅拌30分钟,随后升温至0℃继续搅拌2小时。反应完成后,于减压条件下浓缩反应液,避免加热。向浓缩物中加入乙酸乙酯,所得溶液用饱和氯化铵溶液洗涤。分离有机层,用无水硫酸钠干燥,再次于减压条件下浓缩,避免加热。残余物通过硅胶柱色谱法纯化,洗脱剂为乙酸乙酯/石油醚(1:1,v/v),得到4,6-二氯-1-甲基-1H-吡唑并[3,4-d]嘧啶(6g,78%收率),为白色固体。LC-MS(ES,m/z):203 [M + H]+。
