Uses

2-Methylthiazole-4-carboxamide is a useful reactant for the synthesis of bisthiazole-based histone deacetylase inhibitors.

Step 1. Synthesis of 2-methylthiazole-4-carboxamide (Compound 2A) Ethyl 2-methylthiazole-4-carboxylate (Compound 1, 0.8 g, 4.68 mmol) was mixed with an ethanol solution of ammonia (NH3/EtOH, 10 mL) and the reaction was stirred at 60 °C for 6 days. After completion of the reaction, the mixture was concentrated under reduced pressure to give the crude product. The crude product was purified by column chromatography (eluent: petroleum ether/ethyl acetate=1:1) to afford 2-methylthiazole-4-carboxamide (Compound 2, 0.44 g, 77% yield) as a white solid.
