| Synthesis | The general procedure for the synthesis of 4-iodo-7H-pyrrolo[2,3-d]pyrimidines from 4-chloro-7H-pyrrolo[2,3-d]pyrimidines is as follows: 4-chloro-7H-pyrrolo[2,3-d]pyrimidines were dissolved in 57 wt. % aqueous hydriodic acid. The reaction mixture was stirred at room temperature for 48 h. After completion of the reaction the insoluble solids were removed by filtration. The filtrate was diluted with cold water and the pH was adjusted with aqueous ammonia solution to 8.
